摘  要:目的研究质粒真核表达载体pSVTGF-β1直接注射于瘤体内对荷瘤动物瘤体大小、生存期以及全身细胞免疫功能的影响。方法将艾氏腹水瘤细胞株接种于小鼠左侧腋下,10天后于治疗组动物瘤体中心注射pSVTGF-β1,对照组注射生理盐水。两周后取脾脏检测CTL活性,同时观察肿瘤生长情况及荷瘤小鼠生存时间。结果治疗组动物肿瘤生长速度显著减慢,生存时间显著延长,但两组全身细胞免疫功能无明显差异。结论 TGF-β1基因转移对小鼠实体瘤有一定的治疗作用。Objective To investigate the influence on the tumor growth, life-span and systemic immune func-tion of the tumor bearing mice following direct gene transfer of the plasmid encoding TGF-β1. Methods Ten days after the mice were subcutaneously inoculated with Ehrlich ascites carcinoma cells, the ex-pression vector pSVTGF-β1 was directly injeeted into the middle of the tumor mass, and normal samline was injected into the rats as control group. Two weeks later ,the splenic cells of tumor bearing mice were harvested and the primary cytotoxic T lymphocyte activity was determined. The growth of tumor and the life-span of tumor bearing mice were observed at the same time. Results The growth of the tumor were significantly lower in the treated mice than in the control group. The life-span of the treated group was much longer than that of the control group. There was no difference of the systemic cellular immune function between the two groups. Conclusion TGF-β1 might have a therapeutic effect on the tumor bearing mice.
