摘 要:目的:观察CD44拼接变异体(CD44v)mRNA在胰腺癌组织中的表达及其意义。方法:应用逆转录聚合酶链反应(RT-PCR)及Southern杂交技术对7例正常胰腺组织及27例胰腺癌标本中的CD44vmRNA表达进行研究。结果:7例正常胰腺组织无1例表达CD44vmRNA,胰腺癌组织CD44vmRNA表达率为77.78%(21/27)。统计学分析显示,CD44vmRNA表达与胰腺癌伴随局部淋巴结转移及临床分期呈非常显著相关(P<0.01),与胰腺癌病理分级呈显著相关(P<0.05)。Objective: To observe the expression of metastasis associated splice variants of CD44 (CD44v) mRNA in pancreatic adenocarcinoma and to analyse the relations between expression of CD44v and clinical manifestation in pancreatic adenocarcinoma. Methods: Using reverse transcription polymerase chain reaction (RT PCR) and Southern blot technique, we studied the CD44v mRNA expression in normal pancreatic tissues and pancreatic adenocarcinomas. Results: We found that there was no CD44v mRNA expression in normal pancreatic tissues, whereas 21 of 27 (77.78%) pancreatic adenocarcinomas showed expression of CD44v mRNA. Statistical analysis showed that CD44v mRNA expression in pancreatic adenocarcinoma was related to lymph node metastasis (P<0.01) and to TNM staging (P<0.01), and that the expression was negatively correlated with the extent of histological differentiation (P<0.05). Conclusion: These results suggest that CD44v mRNA expression can be regarded as one of the prognosis factors for the patients with pancreatic adenocarcinoma.
