摘  要:目的研究在抗原提呈过程中HLAⅠ类抗原各相关分子的表达水平及临床意义。方法利用免疫组织化学法(ABC法)对165例肝癌的石蜡组织标本中HLAⅠ类分子的重链、轻链及抗原提呈相关分子calnexin、LMP2等的表达情况进行研究,并分析各表达分子的相关性。结果HLAⅠ类分子(A和B/C位点)的表达为膜型,β_2M的表达为膜浆型,以膜为主,而LMP2和calnexin的表达为浆型。肝癌组织除LMP2分子外,HLAⅠ类各分子均有明显的上调(上调率≥40%),其中以HLAⅠ类分子A位点上调最明显(54%)。肝癌组织中HLAⅠ类各分子之间表达趋势一致。结论肝癌表面HLAⅠ类分子表达增加与抗原提呈过程中多分子的表达增加和协同作用有关,而并非由其中某种单一分子的改变来决定。Objective To investigate the expression of HLA molecules of antigen processing pathway in liver cancer specimens, nethotis The expression of HLA heavy chain, light chain, calnexin, and LMP2 was analyzed by immunohistochemistry ( ABC method) in 165 formalin-fixed,paraffin-embedded liver cancer specimens. Results HLA heavy chain showed membrane staining whereas LMP2 and calnexin had cytoplasmic staining, and β-2m displayed both stainings. HLA heavy chain, light chain, and calnexin were obviously upregulated in most of the liver cancer tissues compared to their normal compartments with the downregulation of LMP2 as an exception. Increased expression of HLA-A was shown in 54% of liver cancer cases, which was the highest of all the molecules investigated. The expression of HLA molecules was correlated with each other and most molecules showed increased expression in the same specimen. Conclusions Increased surface expression of HLA class I complex is due to the upregulation of several molecules of the antigen processing pathway at the same time.
