摘 要:目的探讨外源性补偿嘌呤核苷酸对海洛因依赖大鼠嘌呤核苷酸含量的影响。方法将30只雄性Wistar大鼠随机分为对照组、海洛因组、海洛因+嘌呤核苷酸组(HAG)。各组采用高效液相色谱法检测大鼠脑中GMP、AMP、GTP、ATP含量。结果海洛因组与对照组比较AMP、GTP含量明显降低(P<0.01),GMP、ATP含量改变不明显。补偿嘌呤核苷酸后,AMP含量比海洛因组升高明显(P<0.01)。结论海洛因成瘾大鼠脑中存在嘌呤核苷酸缺失的现象,外源性补偿嘌呤核苷酸能够代偿体内嘌呤核苷酸缺失的状态。Objective To investigate the effect of purine nucleotides compensation on content of purine nucleotides and its possible mechanisms for heroin addiction. Methods Thirty male Wistar rats were randomly divided into there groups ( n = 10) : control group,heroin group,heroin plus AMP and GMP group (HAG).The concentrations of GMP, AMP, GTP and ATP in brain were examined by HPLC. Results The concentrations of AMP and GTP in brain in heroin group were significantly decreased compared with control group ( P 〈 0.01 ). While in HAG group, AMP content was increased when compared with heroin group ( P 〈 0.01 ). Conclusion Exposition of heroin decreased the content of purine nucleotides. Compensation of purine nucleotides can relieve this status.
