摘 要:目的探讨β1整合素表达下调对人结肠癌细胞(HT-29)肝转移的抑制作用。方法采用脂质体将β1整合素反义寡核苷酸(ASODN)转染到HT-29细胞内,逆转录-聚合酶链反应(RT—PCR)和Westernblot检测β1整合素的表达。Transwell侵袭室方法检测HT-29体外侵袭转移能力。裸鼠开腹于脾脏被膜下注射转染的HT-29建立肝转移模型,30d后处死裸鼠,计算肝转移癌的数量与大小,免疫组织化学(SP法)观察转移癌中β1整合素的表达。结果与对照组比较,实验组β1整合素mRNA和蛋白表达水平明显降低(P〈0.05);Transwell体外侵袭实验实验组迁移的细胞数明显减少(P〈0.05);体内裸鼠肝转移实验组转移癌数量(4.00±0.93)个/只、平均体积(10.10±6.50)mm^3/只、平均重量(0.0440±0.0008)g/只、免疫组织化学肝转移癌中β1整合素表达阳性细胞百分率(28.60±3.79)%均明显低于对照组,差异有统计学意义(P〈0.05)。结论β1整合素表达下调对结肠癌肝转移具有抑制作用。Objective To observe the inhibiting effect of down-regulation of integrin-β1 expression on hepatic metastasis of human colon carcinoma cell (HT-29). Methods Integrin-β1 antisense oligodeoxynucleotide (ASODN) was transfected into HT-29 cell by liposome. The expressions of integrin-β1 mRNA and protein were determined by RT-PCR and Western-blot. The migration of HT-29 cells was assayed using transwell cell culture chambers in vitro. The models of hepatic metastasis in nude mice were established by the intrasplenic injection of transfected HT-29 cells. Thirty days later, the number and size of hepatic metastases were calculated, and the expression of integfin-β1 was studied by immunohistochemistry (SP). Results In contrast with the control group,the expression of integrin-β1 mRNA and protein were significantly down-regulated in the experimental group ( P 〈 0.05 ) ; In vitro, the number of migrated HT-29 cells in the experimental group was far less than that of the control group (P 〈0.05 ) ;In vivo,the average number (4.00 ±0.93 )/mouse,volume ( 10.10 ± 6.50) mm3/mouse ,weight (0. 0440 ~0. 0008) g/mouse and positive cell percentage (28.6 ± 3.79) % of hepatic metastases of the experimental group in nude mice were markedly decreased compared with the control group ( P 〈 0.05 ). Conclusion Downregulation of integrin-β1 expression may play a key role in inhibiting hepatic metastasis of human colon carcinoma.
