摘 要:目的:探讨缺氧微环境下单核/巨噬细胞对舌鳞癌细胞吞噬和杀伤作用的影响。方法:从健康人的外周血中分离单核细胞后通过培养将其转化为单核/巨噬细胞;分别在常氧(20%O2)和缺氧(O2<1%)条件下进行单核巨噬细胞和舌鳞癌细胞Tca 8113的混合培养,4 h后MTT法检测单核/巨噬细胞对细胞的杀伤作用。结果:在缺氧微环境下,单核/巨噬细胞对舌癌细胞株的杀伤作用低于常氧环境下(P<0.05)。结论:在缺氧的微环境下,单核/巨噬细胞对舌癌细胞株的吞噬、杀伤作用减弱,但单核/巨噬细胞在肿瘤组织中的作用的具体机制及在肿瘤免疫治疗中的意义还需进一步研究。Objective:To investigate the cytotoxicity of monocytes/macrophages against Tca8113 cells in hypoxic microenvironment in ritro. Methods: Peripheral blood mononuclear cells were extracted from healthy person's blood and cultivated to transform into monocytes/macrophages. The monocytes/macrophages were cultured with Tea8113 cells in hypoxic microenvironment (02 〈 1% ) and in normal mieroeuvironment (20% O2 ). The cytotoxicity of monocytes/macrophages against Tea8113 cell was examined by MTT assay. Results:The cytotoxicity of monocytes/macrophages against Tca8113 cells in hypoxic microenvironment was lower than that in normal environment(P 〈 0.05 ). Conclusion: In hypoxic microenvironment, the cytotoxicity of monocytes/macrophages against Tea8113 cells is decreased. But the mechanism of monocytes/macrophages'function in tumors need further research.
