摘 要:目的:探讨磁流体热疗对小鼠胰腺癌的治疗作用。方法:选择4周龄雌性昆明种小鼠,利用我们前期建株的小鼠胰腺癌细胞系MPC-83建立小鼠胰腺癌皮下肿瘤模型,将0.2mL浓度为1.2g/mL磁流体直接注射到肿瘤内,24h后置入交变磁场中升温至46℃和50℃。作用30min,观察磁流体热疗对荷瘤小鼠的影响及其病理学检查。结果:实验组的肿瘤瘤体中心温度分别迅速升温并稳定在46℃和50℃,而直肠温度保持在30℃~36℃之间。热疗后即刻组织病理学见肿瘤细胞呈凋亡、坏死样改变。热疗后14d实验组小鼠肿瘤体积较各对照组明显缩小,生存期明显延长(P<0.05)。结论:磁流体热疗可以达到杀伤肿瘤细胞的理想温度,能有效抑制小鼠MPC-83胰腺癌的生长,延长生存期,具有较好的治疗效果。Objective: To assess the therapeutic effect of magnetic fluid hyperthermia (MFH) on mouse pancreatic cancer. Methods: Four-week old female Kunming mice were selected in order to establish pancreatic cancer model using the unique mouse pancreatic cancer cell line (MPC-83). At 24 hours after direct injection of magnetic fluid (0.2mL, 1.2g/mL) into the tumor, the mice were respectively heated to 46℃ and 50℃ in an alternating magnetic field for 30 minutes. The therapeutic effect was evaluated by detecting various influences on tumor-bearing mice and pathological examination. Results: The temperature at the tumor center reached 46℃ and 50℃ rapidly, and was kept at the same level by varying the current intensity in magnetic field. In contrast, the temperature of rectal remained below 36℃. Pathological examination showed apoptosis and necorsis of the tumor cells immediately after hyperthermia. Fourteen days after hyperthermia, tumor volume in the experiment groups was significantly lower than that in each control group (P〈0.05). Simultaneously, mice in the experiment group had a longer survival time than those in each control group (P〈0.05). Conclusion: MFH can reach ideal therapeutic temperature and heat tumor tissue specifically, inhibiting the growth of mouse pancreatic cancer and prolonging the survival time of the mice. MFH has good therapeutic efficacy for mouse pancreatic cancer.
