Fas/FasL在人良恶性甲状腺肿瘤中的表达

摘　　要：目的探讨细胞凋亡相关基因Ｆａｓ／ＦａｓＬ在人良、恶性肿瘤中的表达及其意义。方法应用抗Ｆａｓ，ＦａｓＬ多克隆抗体，采用免疫组织化学染色ＳＡＢＣ法，对４４例甲状腺癌，１６例甲状腺瘤和１１例结节性甲状腺肿组织中上述相关抗原的分布规律进行研究。结果Ｆａｓ，ＦａｓＬ在甲状腺癌中的表达阳性率分别为８８．６％（３９／４４）和８４．１（３７／４４）；在甲状腺瘤中的表达阳性率分别为３１．３％（５／１６）和４３．８（７／１６）；在结节性甲状腺肿中的表达阳性率分别为３６．４％（４／１１）和４５．５％（５／１１）。甲状腺癌的表达阳性率明显高于甲状腺瘤和结节性甲状腺肿（Ｐ＜０．００１）。Ｆａｓ、ＦａｓＬ的阳性表达主要位于癌细胞的胞浆内，部分位于胞核和胞膜上，而在甲状腺瘤和结节性甲状腺肿中，则均位于胞浆内和胞膜上。结论Ｆａｓ／ＦａｓＬ基因的表达可能与甲状腺癌的发生及发展相关。Objective:To study the expression and possible significance of apoptosis related gene Fas/FasL in benign and malignant thyroid diseases.Methods:The distributions of Fas/FasL were studied in 44 casess with thyroid carcinoma,16 cases of thyroid adenoma and 11 cases with nodular goiter by immunohistochemical SABC method with polyclonal antibodies against Fas and FasL.Results:The detection rates of Fas and FasL in carcinoma of thyroid were 88.6%(39/44) and 84.1%(37/44) respectively;in thyroid adenoma,those were 31.3%(5/16)、43.8%(7/16),and in nodular goiter,those were 36.4%(4/11)and 45.5%(5/11) respectively.The detection rates of Fas and FasL in carcinoma were much higher than those of thyroid adenoma and nodular goiter( P <0.001).The positive substance was mainly localized in the cytoplasm of cancerous cells,in some cases,it was detected in the nuclei and cell membranne.But in thyroid adenoma and nodular goiter,it was detected in the cytoplasm.Conclusion:The apoptosis related gene Fas/FasL may be involved in development of carcinoma of thyroid.
