摘  要:目的:建立骨转移人乳腺癌细胞株MDA-MB-231BM3及其免疫缺陷小鼠骨转移动物模型。方法:将人乳腺癌细胞株MDA-MB-231改良后,接种于免疫缺陷小鼠左心室形成肿瘤骨转移,在放射性核素示踪下找到裸鼠骨转移病灶,然后切取病变骨组织进行体外培养,获得人乳腺癌骨转移细胞,用上述细胞重复以上体外-体内-体外循环3次,获得骨高转移人乳腺癌细胞株(MDA-MB-231BM3)及乳腺癌骨转移裸鼠模型。并将MDA-MB-231BM3与国外建株的人乳腺癌骨高转移细胞株MDA-MB-231BO在乳腺癌骨转移动物模型建立效率上进行比较。结果:获得以骨转移为主,兼以肺、肾上腺等组织转移的人乳腺癌细胞株MDA-MB-231BM3及其裸鼠骨转移动物模型。结论:MDA-MB-231BM3是以骨转移为主的高转移性人乳腺癌细胞株,该细胞株及其骨转移动物模型为乳腺癌转移的生物学研究提供了一个良好的技术平台。Objective:To establish a human breast cancer cell line (MDA-MB-231BM3) with high bone metastatic potency and its animal model in immune deficient mice. Methods:The human breast cancer ceils MDA-MB-231T were injected into the left cardiac ventricle of nude mice. The metastatic lesions of tumor-bearing mice were imaged by gamma camera. The mice were then sacrificed and the lesions of bone tissues were resected. Metastatic cancer cells in the resected lesions were subjected for culture. This process was re- peated for 3 cycles to obtain a highly metastatic cell subliMe (MDA-MB-231BM3) and its animal model. MDA-MB-231BM3 and MDA- MB-231 BO established in foreign countries were compared on the animal model of bone metastasis of breast cancer. Results:The study, successfully establishes MDA-MB-231 BM3 cell line, which mainly migrates into bone tissues and sometimes migrates into lung and adre- nal gland tissues, etc,and its animal model with bone metastasis in nude mice. Conclusion:MDA-MB-231BM3 has a high bone meta- static potential. The cell line and its animal model provide a good platform for research of the biological behaviors of metastasis of breast cancer.
