摘  要:目的:探讨使用蛋白激酶Cα(protein kinase Cα,PKCα)特异性抑制剂Safingol抑制其活性后对膀胱癌BIU-87细胞增殖能力的影响。方法:应用PKCα抑制剂Safingol、PKCδ特异性抑制剂Rottlerin和PKC激活剂PMA(phorbol-12-myristate-13-acetate)处理人膀胱癌BIU-87细胞,通过MTT法检测细胞增殖能力,RT-PCR及Western印迹法检测立早基因c-fos和c-jun mRNA和蛋白的表达,FCM检验细胞周期的改变。结果:PKCα特异性抑制剂Safingol可明显抑制人膀胱癌BIU-87细胞的增殖并呈剂量依赖性;而PKCδ特异性抑制剂Rottlerin则无此作用,但能提高BIU-87细胞的增殖能力;PKC激活剂PMA则能明显增加BIU-87细胞的增殖能力;FCM检测结果显示,抑制PKCα的活性可使细胞阻滞于G0/G1期;RT-PCR及Western印迹法检测结果表明,c-fos与c-jun的表达与PKCα的活性相关。结论:抑制PKCα活性可明显抑制膀胱癌细胞的增殖能力,其作用机制可能与抑制c-fos和c-jun的表达有关。Objective:To investigate the inhibitory effect of of protein kinase Cα(PKCα) activity by its specific inhibitor Satin- gol on bladder cancer cell proliferation. Methods:The bladder cancer BIU-87 cells were treated with PKCα specific inhibitor Safingol, PKC8 specific inhibitor Rottlerin, and PKC activator phorbol-12-myristate-13-acetate (PMA) , respectively. Cell proliferation was de- termined by MTF assay. The expressions of c-fos and c-jun mRNA and protein were detected by RT-PCR and Western blotting, respec- tively. Cell cycle was detected by flow cytometry. Results : The proliferation of bladder cancer ceils was significantly inhibited by PKCα specific inhibitor Safingol in a concentration-dependent, while PKCδ special inhibitor Rottlerin had no effects but could enhance the ca- pability of cell proliferation. PKC activator PMA significantly enhanced the proliferation potency of BIU-87 cells. FCM analysis showed that inhibition of PKCα activity arrested ceils at G0/G1 phase. RT-PCR and Western blotting showed that expressions of c-los and c-jun were positively related with PKCα activity. Conclusion:Bladder cancer BIU-87 cell proliferation can be inhibited by PKCα specific in- hibitor, which may be related to inhibition of c-fos and c-jun expressions.
