基质固相分散-液相色谱法测定血豆腐中的激素

摘  要:采用基质固相分散样品前处理方法,高效液相色谱同时测定血豆腐中雌三醇、诺龙、甲基睾丸酮、己烯雌酚4种激素。考察了分散剂种类、样品与分散剂质量比、洗脱剂及洗脱体积对测定结果的影响。色谱条件为:DikmaC18柱(5μm,250 mm×4.6 mm i.d.),流动相为V(甲醇)∶V(四氢呋喃)∶V(水)=60∶10∶30。4组分线性相关系数为0.999 8-0.999 9。样品检出限为0.025-0.1μg/g(S/N=3),回收率83.0%-94.0%。Matrix solid phase dispersion - high performance liquid chromatographic (MSPD-HPLC) method was used for the simultaneous determination of four hormones in the blood beam curd sample, including estriol, norethindrone, methyltestosteron and diethylstilbestrol. Parameters such as the kind of dispersant, the weight ratio of dispersant and sample, the kind and volume of eluent were studied and optimized. Dikma C18 column (5μm, 250 mm × 4.6 mm i. d. ) was used for separation of the target compounds. Mobil phase was 60: 10: 30(v/v/v) methanol: tetra-hydrofuran: water. The flow rate was 1.0 mL/min and the wavelengths of UV detector were 280 nm and 241 nm. The linear correlation coefficient of the four hormones were 0. 999 8 - 0. 999 9. The detection limits of the four hormones were 0. 025 -0.1μg/g. The average recovery of the compounds ranged from 82.9% to 93.7%.
