摘 要:目的观察不同痫性发作次数对戊四氮(PTZ)诱导的癫痫大鼠空间学习记忆功能的影响及海马部位细胞外信号调节激酶1/2(ERK1/2)mRNA及蛋白水平的变化。方法PTZ腹腔注射诱导慢性癫痫模型,点燃后根据痫性发作次数分为不同亚组(痫性发作1次、5次、10次、14次组),采用Morris水迷宫检测大鼠行为学变化,采用反转录聚合酶链反应(RT-PCR)及免疫印迹WesternBlot的方法检测大鼠海马ERK1/2mRNA和蛋白水平的变化。结果癫痫组大鼠空间学习记忆能力受损;痫性发作1次、5次组大鼠海马部位ERK1/2mRNA水平较对照组下降(P<0.05),而10次、14次组大鼠海马部位ERK1/2mRNA水平较1次有所升高(P<0.05),且高于对照组大鼠水平(P<0.05)。各组大鼠海马部位总ERK1/2和P-ERK1/2的表达差异无统计学意义(P>0.05)。结论随痫性发作次数增加,癫痫大鼠空间学习能力受损明显,其海马部位ERK1/2mRNA水平存在一个动态变化过程,癫痫后认知功能损害的机制可能与ERK信号通路有关。Objective To explore the relationship between spatial learning and times of seizures and the expression of extracellular signal - regulated kinase1/2 ( ERK1/2 ) in hippocampus of pentylenetetrazol - kindled rats. Methods After being kindled successfully, rats were divided into groups accroding to times of seizures. Their behaviors were afterwards evaluated through Morris water maze and the expression of ERK1/2 in hippocampus was determined by methods of RT - PCR and Western blot. Results Expression of ERK1/2 mRNA decreased in rats with 1 seizures and with 5 seizures (P 〈 0. 05 ), and increased in those with 10 seizures and 14 seizures (P 〈0. 05). There was no significant difference between groups of total ERK and P - ERK1/2 (P 〉 0. 05 ). Conclusion Impaired spatial learning is related with times of seizures, its mechenism possiblely related with the ERK signaling pathway.
