摘 要:目的探讨血小板因子-4(platelet factor-4,PF-4)对脑胶质瘤细胞Ca2+内流的影响。方法脑胶质瘤细胞U251传代培养,分别加入浓度为0(对照组)、900 ng/ml、1200 ng/ml、1500 ng/ml的PF-4,继续培养24 h。利用LSCM测定经Fluo-3标记的脑胶质瘤细胞内Ca2+荧光强度。结果对照组脑胶质瘤细胞内Ca2+荧光强度为24.21±2.36;实验组脑胶质瘤细胞内Ca2+荧光强度分别为39.35±3.31,52.39±3.26,65.56±2.81。经单因素方差分析,各组间差异P<0.05。结论PF-4对脑胶质瘤细胞Ca2+内流有明显的促进作用。Objective: To investigate the influence of PF-4 on calcium flow of human glioma cells.Methods: Human gliomas(U251) were subcultured and the PF-4 concentration of 0 ng/ml(control group)、900ng/ml、1200ng/ml、1500ng/ml were given separately.After the culture of 24 hours,intracellular free Ca2+ fluorescence intensity of Fluo-3-labeled glioma cell was measured by LSCM.Results: The fluorescence intensity of control group was 24.21±2.36 and that of experimental groups were respectively 39.35±3.31,52.39±3.26 and 65.56±2.81.There was a significance by one-way ANOVA.Conclusion: PF-4 can promote the flow of calcium of human glioma cells.
