摘  要:测定了新疆紫草种子萌发过程中过氧化物酶(POD)活性及酚类物质含量的变化,考察了其对新疆紫草种子萌发的影响。结果表明,新疆紫草种子在萌发过程中,随POD活性的增强,发芽率逐渐升高,POD活性增强促进新疆紫草种子的萌发。福林试剂与10NNa2CO3之比为2:1(V:V),反应温度为25℃,反应时间为45min,体积为0.5~3.0mL时与其吸光度值呈良好的线性关系,建立了酚类物质的提取及测定方法。总酚含量随发芽进程呈先降低后升高趋势,酚类物质的合成和积累抑制新疆紫草种子萌发。This study determined the course of seed germination of Arnebia euchroma in seed coat, embryo Peroxidase (POD) activity and changes in polyphenol content, examined their impact on Arnebia euchroma seed germination. The results showed that the germination percentage of seeds in Arnebia euchroma (Royle) Johnst gradually was increased with the rising of POD activity during its germination process.In the Folin reagent,the ratio of nutgall acid to 10% Na2CO3 was two to one (V : V). When the reaction temperature was 25℃ ,reaction time was 45 min. When the volume was 0.5-3.0 mL,its absorbance value was a good linear relationship. The method for extracting and determining phenolic substances was established. In addition,the polyphenol content first was reduced then increased with the proceeding of germination. The synthesis and accumulation of phenolic substances inhibited the germination percentage of Arnebia euchroma (Royle) Johns.
