摘 要:背景与目的:TEIF基因是本室新近发现的一种调节端粒酶hTERT基因表达的转录因子,在多种肿瘤组织中均有表达。本研究观察TEIF基因在结直肠肿瘤组织中的表达及其与中心体改变的关系。方法:通过原核表达TEIF蛋白免疫制备多克隆抗体,经免疫印迹鉴定后,对10例结直肠黏膜正常组织、30例结直肠癌和54例结直肠腺瘤(Ⅰ级18例,Ⅱ级17例,Ⅲ级19例)标本行TEIF蛋白免疫组化检测及γ-tubulin免疫荧光染色检测。结果:免疫组化结果显示正常组中TEIF蛋白的表达率(10.0%)与肿瘤各组之间(恶性肿瘤组表达率100%,良性肿瘤组98.1%)的差异有统计学意义(P<0.01),恶性肿瘤组与良性肿瘤组之间的差异无统计学意义(P>0.05)。癌组织组中TEIF强阳性率(≥++)(100%)均显著高于腺瘤组(61.1%)及正常组(0%)(均为P<0.001);腺瘤Ⅰ级(11.1%)与腺瘤Ⅱ级(76.5%)及Ⅲ级(94.7%)组织中TEIF强阳性率之间的差异有统计学意义(P<0.05),而腺瘤Ⅲ级与腺瘤Ⅱ级间的差异无统计学意义(P>0.05)。免疫荧光结果显示结直肠癌组中心体扩增阳性率(100%)显著高于正常组(0)及腺瘤组(64.8%)(均为P<0.01);腺瘤Ⅰ级(44.4%)与腺瘤Ⅲ级(79.0%)中心体扩增阳性率之间的差异有统计学意义(P<0.05),而腺瘤Ⅰ级与腺瘤Ⅱ级(70.6%)、腺瘤Ⅱ级与腺瘤Ⅲ级之间的差异无统计学意义(P>0.05)。结论:TEIF蛋白及中心体扩增常见于结直肠肿瘤中,其表达水平与肿瘤的组织学分级及恶性程度相关。TEIF蛋白表达与中心体扩增呈高度的正相关。Background and Objective. Telomerase transcriptional elementsinteracting factor (TEIF) gene found recently by our research group is a transcription factor of a kind of human telomerase reverse transcriptase (hTERT) gene, and expresses in many kinds of tumor tissues. This study was to evaluate the expression of TEIF protein in human colorectal tumors and to explore its correlation with centrosome abnormality. Methods: The expression of TEIF in 10 specimens of normal intestinal mucosa tissue, 30 specimens of colorectal cancer, and 54 specimens of colorectal adenoma was detected by immunohistochemistry. The expression of gamma-tubulin was detected by immunofiuorescence. Results. Immunohistochemistry results showed that the differences of TEIF protein expression between the normal group and each tumor groups were statistically significant (P〈0.01), and the difference of TEIF protein expression between the malignant tumor group and the benign group was not significant (P〉0.05). TEIF strong positive rate (≥++) was significantly higher in the carcinoma group than in the adenoma group or normal group (all P〈0.001); the differences of TEIF strong positive rate between Grade I adenoma and Grade Ⅱ or Ⅲ adenoma were statistically significant (P〈0.05), and the difference of TEIF strong positive rate between Grade Ⅱ adenoma and Grade Ⅲ adenoma was not statistically significant (P〉0.05). Immunofluorescence results showed that centrosome amplification-positive rate was significantly higher in the colorectal cancer group than in the normal group or the adenoma group (both P〈0.01); the difference of the centrosome amplification positive rate between Grade I adenoma and Grade III adenoma was statistically significant (P〈0.05), and the differences of the centrosome amplification positive rate between Grade II adenoma and Grade I or III adenoma were statistically significant (P〉0.05). Conclusions: TEIF protein and centrosome amplification is commonly found
