摘  要:目的探讨血管内皮生长因子(VEGF)在血卟啉衍生物(HPD)加光动力作用(PDT)诱导人胰腺癌细胞PANC-1凋亡中的表达。方法应用流式细胞术检测细胞凋亡率,免疫组化染色法检测VEGF蛋白表达,RT-PCR检测基因表达水平。结果HPD+PDT实验组人胰腺癌细胞PANC-1凋亡率达(36.40±4.21)%,明显高于单纯HPD或PDT及空白三个对照组(P<0.05),VEGF蛋白和基因的表达均显著低于三个对照组(P<0.05),三个对照组间细胞凋亡、VEGF蛋白和基因表达率差异均无统计学意义。结论HPD+PDT可诱导人胰腺癌细胞PANC-1凋亡和抑制VEGF表达,进而抑制肿瘤血管生长。Objective To evaluate the effect of photodynamic therapy on vascular endothelial growth factor(VEGF) expression in pancreatic carcinoma cell PANC-1. Methods The apoptosis of human pancreatic carcinoma cell PANC-1 was detected by flow cytometry with PI and FITC staining. The expression of VEGF of cell PANC-1 was determined by immunocytoehemistry and RT-PCR assay, respectively. Results The apoptosis rate of pancreatic carcinoma cell PANC-1 after photodynamic therapy with hematoporphyrin was ( 36.40±4. 21 ) %, which was significantly higher than that in the controls (P〈 0. 05). The expression of VEGF and VEGF mRNA in PANC-1 matoporphyrin group was lower than that in the controls (P〈0.05) while no difference between the controls. Conclusion The expression of VEGF in pancreatic carcinoma cell PANC-1 can be inhibited by photodynamic therapy.
