摘  要:目的:利用食源性肥胖(DIO)大鼠模型研究肥胖对脾脏CD4+和CD8+T淋巴细胞含量的影响,并探讨其是否与脾细胞内JAK/STAT通路相关联。方法:(1)通过高能饲料喂养建立DIO大鼠模型。(2)利用流式细胞术检测大鼠脾脏CD4+和CD8+T淋巴细胞含量。(3)利用Western blot检测大鼠淋巴细胞长型的leptin受体(Ob-Rb)以及活化的STAT3蛋白的表达。结果:(1)高能饲料喂养的大鼠体重高于普通饲料喂养大鼠,发育为DIO大鼠。(2)DIO大鼠脾脏CD8+T淋巴细胞含量高于肥胖抵抗(DR)组以及对照组的大鼠,CD4+/CD8+细胞比值低于DR组以及对照组的大鼠。(3)各组大鼠脾脏淋巴细胞Ob-Rb以及活化的STAT3蛋白的表达没有统计学差异。结论:DIO大鼠脾脏CD8+T淋巴细胞含量高于非肥胖大鼠,CD4+/CD8+细胞比值低于非肥胖大鼠,其处于免疫抑制状态;这一变化与DIO大鼠脾脏淋巴细胞的Ob-Rb-STAT3信号转导通路并没有直接的关系。Objective: To study the influence of obesity on splenic CD4^+ and CD8^+ T-lymphocytes, and observe the relationship between obesity and the JAK/STAT pathway of splenic lymphocyte. Methods: (1) DIO rat model was established by feeding high-fat diet. (2) Flow-cytometry was used to detect splenic CD4^+ and CD8^+ T-lymphocytes of rats. (3) Western blot was used to study the expression of the long signaling isoform of leptin receptor (Ob-Rb) and phospho-stat3 (Tyr705). Results: After high-fat diet feeding, rats developed diet-induced obesity; body weight of DIO rats was greater than that of the control rats. The number of DIO rats' splenic CD8^+ T-lymphocytes was increased significant than that of DR and lean rats', and DIO rats' ratio of splenic CD4^+/CD8^+ lymphocytes was decreased than that of DR and lean rats'. Expression of Ob-Rb and phospho-stat3 (Tyr705) had no difference among control, DR and DIO group (P〉0.05). Conclusion: DIO rat has more splenic CD8^+ T-lymphocytes and less ratio of splenic CD4^+/CD8^+ lymphocytes, DIO rat exhibits an immune inhibition response,and there is no relation between this change and Ob-Rb- STAT3-signaling transduction pathway.
