摘 要:目的:探讨冷冻对小鼠前列腺癌血管生成及肿瘤细胞增殖的影响。方法:建立小鼠前列腺癌肿瘤模型,随机分为对照组和冷冻组,冷冻组实施氩氦冷冻治疗术。两组分别在不同时间点用免疫组化方法检测肿瘤组织的VEGF、Ki67蛋白表达。结果:对照组各时间点VEGF、Ki67蛋白表达无明显变化(P>0.05)。冷冻组在冷冻后VEGF、Ki67蛋白表达均出现下降,低峰值出现在第3天,然后缓慢回升,与对照组相比,有显著性差异(P<0.01);冷冻组间VEGF、Ki67经统计分析有显著性差异(P<0.05)。冷冻后前列腺癌VEGF蛋白表达水平与Ki67呈显著正相关(r=0.6921,P<0.01)。结论:冷冻可明显减低肿瘤VEGF、Ki67的表达水平,可起到抑制肿瘤生长作用。Objective: To explore the effects of cryotherapy on the angiogenesis and cell proliferation in prostate cancer of mice. Methods: Mouse experimental models with prostate cancer were established. Experimental mice were randomly divided into control group and freezing group. Ki67, VEGF protein expression were detected by immunohistochemical staining at different time after the freezing. Results: There were no change in Ki67, VEGF protein expressions at different time in control group (P〉0.05), Ki67, VEGF protein expressions in freezing group were decreased.The lowest value was detected at the third day after freezing,the value increased slowly after that (P〈0.01).There were significant changes in VEGF, Ki67 protein expressions at different time in freezing group (P〈0.05). Correlation analysis showed that VEGF protein expression was positively correlated with Ki67(r=0.6921,P〈0.01). Conclusion: Cryotherapy can considerably reduce VEGF and Ki67 protein expression,thus inhibiting the growth of prostate cancer in mice.
