摘 要:目的:探讨3种细胞因子单一、协同以及与化疗药物顺铂(DDP)的联合作用杀伤肝癌SMMC-7221细胞、宫颈癌Hela细胞株的影响。方法:应用四甲基偶氮唑盐比色法(MTT比色法)、检测与化疗药物DDP单独或联合应用组及药物作用的12 h与24 h的光密度值(OD),计算杀伤率,以台盼蓝拒染计算杀伤率对照。结果:单独应用3种细胞因子时,IFN-a、TNF-a对肝癌细胞、宫颈癌Hela细胞毒作用,IL-2对肝癌细胞、宫颈癌Hela细胞则细胞毒作用不明显。IFN-a、IL-2、TNF-a可以增强DDP的抑瘤作用,与DDP单独作用具有显著性差异(P<0.01)。IFN-a、IL-2、TNF-a联合作用具有协同作用,均与单一应用具有显著性差异(P<0.01)。细胞因子均可以增强顺铂(DDP)的抑瘤作用并与其作用的先后及时间具有依赖关系。结论:MTT比色法为临床肿瘤化疗的药物敏感性检测提供了简便、快速的方法。不同类型的肿瘤有着不同的药敏谱。干扰素-a(IFN-a)、肿瘤坏死因子(TNF-a)、白细胞介素-2(IL-2)与DDP联合具有协同作用,为肝癌、宫颈癌病人化疗提供了理想的参考方案。Objective: To investigate the effects of three alone and in combination chemicals with DDP on Liver cancer cell or Hela cell line. Methods: Chemosensitivity of cells to doxorubicin was analysed using the MTT Assay 12h and 24h after using three cell genes alone and in combination chemicals with DDP chemotherapy drug, and compared using typanlan examined. Restults The results showed that there were significant diffenrnces effects on the Liver cancer cell or Hela cell llne in the inhibition rates of the between rhIFN - a group, rhTNF - a group and negative control group. IL - 2 had not strong activity of antitumor, rhTNF - a and rhIFN - a had a strong activity of antitumor in vitro and good dose - effect relationship compared with using DDP alone (P 〈0. 01 ) . rhIFN - a, rhIL -2, rhTNF - a were significant diffenrnces effects compared with using one alone group (P 〈0. 01) . rhlFN - a, rhIL -2, rhTNF - a with DDP had synergistic enhancement of antitumor activity in vitro and depend on time. Conclusion The MTr method is very rapid and simple to perform. Chemosensitivity spectra vary with tumor types. It can be applied to clinical drug sensitivity testing of human tumors and the results of this chemosensitivity will provide the reference or ideal plan for the chemotherapy of cervical carcinoma patients.
