摘  要:目的研究ATX基因短发夹状RNA(shRNA)质粒表达载体对人MHCC-97肝癌细胞ATX基因表达和增殖、运动能力的抑制作用。方法应用RT-PCR检测转染后肝癌细胞ATX mRNA的表达,同时应用MTT、细胞运动、侵袭实验检测其对人MHCC-97肝癌细胞增殖、侵袭、运动能力的影响。结果ATX-shRNA表达载体可以有效的抑制人MHCC-97肝癌细胞中ATX基因的表达,使ATX基因的mRNA表达量显著下降,以转染后第四天下降最明显,并且转染后细胞增殖、侵袭、运动能力均减弱。结论ATX-shRNA表达载体能有效地抑制ATX基因表达。Objective To investigate the short hairpin RNA (shRNA)expression plasmid of ATX on inhibiting ATX mRNA expression and cell proliferation and motility in human hepatocellular carcinoma cells. Methods The ATX mRNA expression was measured by semi-quantity RT-PCR, meanwhile,The cell viability and activity were determined by MTT assay and the changes of invasioness and cell motility were investigated by transwell chamber system. Results ATX mRNA expression was efficiently suppressed by plasmid expressing shRNA targeting ATX, The best inhibition of expression of ATX gene was at the fourth day using pSilencer-ATX. Conclusion ATX-shRNA expression plasmid can significantly knock down ATX gene expression in MHCC-97 cells.
