摘 要:目的:探讨细胞凋亡相关基因Bcl-2、Bax、Bak mRNA在子痫前期患者胎盘组织中的表达及其意义。方法:采用RT-PCR方法测定36例正常足月妊娠妇女(A组)、24例轻度子痫前期患者(B组)和17例重度子痫前期患者(C组)胎盘组织中Bcl-2、Bax、Bak mRNA表达水平。结果:(1)与B组(1.563±0.398)和A组(1.723±0.300)相比,C组Bcl-2 mRNA表达水平(0.642±0.288)明显降低(q=12.41,P<0.05;q=15.69,P<0.05),B组与A组之间无明显差异(q=2.6,P>0.05)。(2)与A组(0.347±0.191)相比,B、C组BaxmRNA表达水平(分别为0.745±0.260、1.335±0.308)明显升高(q=8.79,P<0.05;q=19.54,P<0.05),C组较B组升高更为显著(q=10.83,P<0.05)。(3)与B组(0.875±0.322)和A组(0.809±0.381)相比,C组Bak mRNA表达水平(2.197±0.546)明显升高(q=14.52,P<0.05;q=16.42,P<0.05),B组与A组之间无明显差异(q=0.87,P>0.05)。结论:细胞凋亡相关基因Bcl-2、Bax、Bak mRNA在子痫前期患者胎盘组织中的差异表达参与了子痫前期的病理生理过程。Objectives To investigate the expression and significance of apoptotic correlated gene Bcl-2, Bax and Bak mRNA in preeclampsia placenta. Methods The levels of Bcl-2, Bax and Bak mRNA expression in placenta of 36 healthy term pregnant women (group A), 24 patient with mild preeclampsia (group B), and 17 patients with severe preeclampsia ( group C) were examined with RT-PCR. Results ( 1 ) Expression of Bcl- 2 mRNA in placental tissues in group C (0. 642± 0.288 ) was significantly lower than that in group B ( 1. 563 ±0.398 ) and group A ( 1. 723±0. 300) ( q = 12.41, P 〈 0.05 ; q = 15.69, P 〈 0.05 ), while no statistical differences were observed between group B and group A ( q = 2.6, P 〉 0.05 ). ( 2 ) Expression of Bax mRNA in placental tissues in group B ( 0. 745 ±0. 260 ) and group C ( 1. 335 ± 0. 308 ) were significantly higher than that in group A (0. 347 ±0.191 ) ( q = 8.79, P 〈 0.05 ; q = 19.54, P 〈 0.05 ), and the level of Bax mRNA in placental tissues in group C was higher than that in group B ( q = 10.83, P 〈 0.05 ). ( 3 ) Expression of Bak mRNA in placental tissues in group C (2. 197 ±0. 546 ) was significantly higher than that in group B (0. 875 ±0. 322) and group A (0.809 ±0.381 ) (q = 14.52,P 〈 0.05 ;q = 16.42, P 〈 0.05 ), while no statistical differences were observed between group B and group A ( q = 0.87,P 〉 0.05 ). Conclusion The differential expression of apoptotic correlated gene Bcl-2 ,Bax and Bak mRNA in preeclampsia placenta play an important role in the physiological and pathological process of preeclampsia.
