摘  要:目的:观察大鼠弥漫性轴索损伤(DAI)后β-淀粉样前体蛋白(-βAPP)表达的变化规律。方法:实验组75只SD大鼠按照打击负荷伤模型致伤,于伤后4 h、8 h、24 h、72 h和1周时分批处死,取大鼠脑组织切片行β-APP免疫组织化学染色,计算并比较大鼠外伤后各时间点脑组织-βAPP免疫组化阳性染色面积百分比、平均光密度值及阳性细胞数。另取8只大鼠作为对照组,处死后采用同样方法观察脑组织-βAPP的表达,并与实验组作比较。结果:与对照组比较,实验组各时间点脑组织-βAPP阳性免疫反应明显增强且范围明显增大,半定量分析显示差异有统计学意义(P<0.05),并于外伤后72 h最强,1周时开始减弱。结论:大鼠DAI后脑组织-βAPP的表达持续增强,于72 h达顶峰,1周时开始减弱。Objective:To investigate the expression changes of β-amyloid precursor protein(β-APP) after diffuse axonal injury(DAI) in rats.Methods: The Marmarou's model was applied to induce DAI in 75 SD rats and another 8 rats were served as normal control group.At assigned time points(4,8,24,72 h,and 1W post-trauma),the rats were killed and prepared for histological analysis by using a rabbit antibody against β-APP.The expression changes of β-APP at different stages post-trauma were evaluated according to immunoreactivity of brain tissues.Results: β-APP immunohistochemistry showed that the immunoreactivity of brain tissues of the experimental group was significantly enhanced when compared with the control group.Semi-quantitative analysis further revealed that the immunoreactivity between experimental and control group had significant difference(P0.05).It was shown that the immunoreactivity was strongest at 72 h and declined at 1w after injury.Conclusion: The β-APP expression increased after DAI in rats,reached the peak at 72 h and declined at 1 week.
