摘 要:产乙醇杆菌YUAN-3^T是以糖蜜废水为底物分离出的具有良好自凝集现象的高效产氢菌株,在连续流反应器中减少了产氢细菌的流失,客观上起到了固定化的作用,保证了反应器内产氢细菌的高浓度,提高了糖蜜的利用率,降低了制氢成本,进一步加快了发酵法生物制氢技术的工业化进程。为了提高菌体产氢能力,并逐步阐明YUAN-3^T在包括糖蜜在内的多种基质中发酵的活力影响因素,通过发酵罐静态试验及优化的氢化酶活性检测方法,分析了pH对YUAN-3^T产氢能力及氢化酶活性相关性。结果表明,YUAN-3^T的产氢能力对pH变化极为敏感,生物量在第9h即pH开始快速下降时进入生长对数期,pH降为4时发酵缓慢,产气基本停止,生物量在24h达到最大值后开始减少。氢化酶活性随代谢过程迅速升高并在pH4.5时达到最大值后下降。High efficient H2 - producing bacterium E. harbinense YUAN - 3T is an auto - aggregative strain which is isolated from continuous stirred - tank reactor for the molasses wastewater as substrate. Reactor can ensure the hydrogen - producing bacteria in the high concentration to increase the utilization of molasses. In order to improve the hydrogen production efficiency of the bacteria, illustrate the effecting factors of the enzyme activity of YUAN - 3T in multiple culture system involving molasses wastewater, and speed up the fermentation technology further, the relationship between pH value and the hydrogenases activity of YUAN - 3 was analyzed using batch test in the present study. Results show that, YUAN - 3 is extremely sensitive to pH value for its hydrogen yield. The biomass drops at the beginning of logarithmic phase into the growth as the pH falls rapidly at the ninth hour. The fermentation goes slowly when pH reaches 4, and the hydrogen - production stops at the same time. The biomass decreases after reaching a maximum at 24h along with the fermentation process. The activity of Hydrogenases increases rapidly with a maximum when pH reaches 4.5.
