摘  要:目的探讨大鼠侧脑室注射链脲霉素模型与散发性阿尔茨海默病(sporadic Alzheimer's disease,SAD)的相关性。方法S-D大鼠分别于第1天和第3天双侧侧脑室注射链脲酶素(STZ)建立模型。21d后检测琥珀酸脱氢酶(succinate dehydrogenase,SDH)、细胞色素C氧化酶(cytochrome-oxydase,CCO)和Na+-K+-ATPase、Ca2+-Mg2+-ATPase活性;高效液相法检测ATP浓度;免疫组化法检测Aβ1-40、Aβ1-42阳性细胞在海马区表达。结果STZ组大鼠脑内SDH、CCO和Na+-K+-ATPase、Ca2+-Mg2+-ATPase活性及ATP浓度与NS组相比均有显著降低。STZ组大鼠海马区Aβ1-40、Aβ1-42阳性细胞较NS组显著增多。结论大鼠侧脑室注射链脲霉素可导致ATP减少及脑内Aβ1-40、Aβ1-42表达增多等与SAD相类似的病理变化。Objective To testify whether intracerebroventricular(icv) injection of STZ can cause energy deficiency with increased Aβ1-40,Aβ1-42 positive cells in rat brain that similar to changes of sporadic Alzheimer's disease(SAD).Methods S-D rats were divided into groups to receive icv injection of STZ or normal saline(NS) bilaterally at 1st and 3rd day.After 21 days of operation,activities of succinate dehydrogenase(SDH),cytochrome-oxydase(CCO) and Na^+-K^+-ATPase,Ca^2+-Mg^2+-ATPase were measured.ATP levels of groups were detected by high performance liquid chromatography(HPLC) method.Immunohistochemical method was used to detect Aβ1-40 and Aβ1-42 positive cells in hippocampus of rat brain.Results Rats received icv injection of STZ bilaterally showed decreased ATP concentration and increased positive cells of Aβ1-40,Aβ1-42 in the brain.Conclusions Introcerebroventricular injection of STZ can cause similar changes to pathogenetic disorders of SAD.
