摘  要:目的:观察125Ⅰ粒子在瘤内放疗半径。方法:采用BALB/C裸鼠腺样囊性癌(adenoidcysticcarci-noma,ACC)模型10只,实验组和对照组各5只,分别于肿瘤中央植入活度为10.2MBq和无活度的空心125Ⅰ粒子1枚,14d后断颈处死裸鼠,制作肿瘤组织标本,行透射电镜及常规病理学染色检测肿瘤坏死凋亡情况并记录肿瘤出现坏死凋亡改变距离125I粒子的最大半径。结果:近125I粒子绝大部分癌细胞坏死。多数癌细胞内形成坏死腔,将残存细胞质挤向周边;细胞核碎裂部分融解,染色质呈斑块状,胞核胞质难以分辨,细胞质内结构模糊不清,细胞表面微绒毛极少。125I粒子最大有效半径处ACC细胞由正常癌细胞到凋亡坏死逐渐过渡的过程,可见癌细胞呈不同程度的损伤,包括凋亡细胞(体积小于正常癌细胞,核膜胞膜清晰)、坏死细胞及未受损伤的癌细胞混杂。125I粒子最大有效治疗半径为6.4±0.7mm。结论:125I粒子可以导致癌细胞的坏死凋亡,有效治疗半径为6.4±0.7mm。AIM: To observe the radiation radius of ^125I particles in the tumor.METHODS: Ten BALB/C nude mouse model of adenoid cystic carcinoma were divided into test group and control group medially. One ^125Iodine seed ( radioactivity 0, 6mci) was implanted into the middle of each tumor in test group. While each non-radioactive ^125Iodine seed was implanted into the tumor in control group. On the 14^th day after seed implantation, tumor tissues underwent routine histopathologic examinatinon and transmission electron microscopy to observe tumor cell apoptosis and necrosis and measure the maximum radius between changed cell and ^125Iodine seed. RESULTS: Near to ^125Iodine seed, transmission electron microscope showed that there were necrotic cells absolutely. Low power lens showed that necrotic cavity was formed in tumor cells, which made remanent cytoplasm limbic. On high power lens, necrosis was apparent which was characterized by nuclear fragmentation and deliquescing, chromoplasm flocculation, the obscure and unrecognizable structure of cytoplasm and nucleus. Less fluff was on the surface of membrane. Results of transmission electron microscope showed the gradual course from normal cells to apoptosis and necrosis. It appeared the tumor cells injury to different extent, including apoptosis cells(with clear membranes, less volume than that of normal tumor cells), necrotic cells and uninjuried tumor cells. Maximum effective radius of ^125I seed was 6.4±0.7mm.CONCLUSION: ^125Iodine seed leads to apoptosis and necrosis of tumor cells. According to cellular changes under microscopes, clinicians can identify effective radius of ^125Iodine seed implantation, and it will provide a reference for the clinical design of the ^125Iodine seed implantation.
