摘 要:目的检测正常鼻咽组织以及鼻咽非角化性癌癌细胞Fas的表达,探讨其表达与癌细胞增生和凋亡的关系。方法收集中山大学附属第一医院病理科37例未治疗的非角化性鼻咽癌活检组织标本。应用免疫组化法检测鼻咽癌组织中Fas和癌细胞PCNA的表达;应用原位细胞凋亡检测法(TUNEL)计数癌细胞的凋亡(Apoptosis Count,AC)。结果正常鼻咽粘膜表面及隐窝的假复层纤毛柱状上皮细胞、鳞状化生上皮、部分小涎腺的导管上皮细胞的胞膜和胞浆Fas阳性表达。45.9%(17/37)的鼻咽非角化性癌的少数癌细胞Fas弱阳性。癌细胞表达Fas与癌细胞凋亡计数AC无关(P>0.05)。Fas阴性病例癌细胞增生指标显著高于Fas阳性病例(P<0.01)。结论Fas对鼻咽非角化性癌癌细胞的自发性凋亡不起主导作用,鼻咽非角化性癌细胞大多处于细胞增生周期是导致癌细胞自发性凋亡非Fas途径的主要原因之一。Objective To detect the expression of Fas in normal nasopharyngeal tissue and non-keratinizing carcinoma (NKC) cells and investigate the relationship between Fas expression and neoplastic cell proliferation, apoptosis. Methods 37 untreated nasopharyngeal non-keratinizing carcinoma biopsies were collected from the Department of Pathology, The First Affiliate Hospital, Sun Yat-sen University. Immunohistochemistry was used to detect the expression of Fas and PCNA. TUNEL method was used for counting the number of apoptotic cell indicated as AC (Apoptosis Counting). Results The pseudostratified ciliated columnar epithelium and metaplastic squamous epithelium, as well as part of ductal cells of minor salivary gland had detectable membranous expression of Fas. 17 out of 37(45.9%) NKC samples was found Fas weak expression only in part of neoplastic cells. There was no correlation between AC of Fas expression in NKC (P〉 0.05). How- ever, the neoplastic proliferation index in Fas negative tissues was significantly higher than that of Fas poitive tissues (P〈 0.01). Conclusions The Fas might be not a major pathway involved in NKC cell spontaneous apoptosis. The most of NKC cells underwent proliferating cell cycle might be an important factor to induce cell apoptosis through Fas-independent pathway.
