mIL-2分泌型小鼠黑色素瘤细胞诱导的细胞免疫应答

摘　　要：用ｍＩＬ２分泌型小鼠黑色素瘤细胞（ｍＩＬ２＋Ｂ１６）植入同种小鼠（Ｃ５７ＢＬ／６）腹腔内诱导腹腔淋巴细胞（ＰＥＬ）。对ｍＩＬ２＋Ｂ１６及其亲代Ｂ１６细胞在不同天数内诱导的ＰＥＬ的数量、ＰＥＬ组成成分、对靶细胞的杀伤百分率等几个方面的特点进行了动态分析和比较，结果显示：不同诱导天数的ＰＥＬ之间，以上三个特点存在明显的动态变化规律，而ｍＩＬ２＋Ｂ１６和Ｂ１６的ＰＥＬ之间的这种动态变化又存在明显的差别。在ＰＥＬ的杀伤百分率上，ｍＩＬ２＋Ｂ１６ＰＥＬ比Ｂ１６ＰＥＬ却高一倍以上。电镜下观察ＰＥＬ胞浆中的特殊颗粒（含Ｐｅｒｆｏｒｉｎ，Ｇｒａｎｚｙｍｅｓ）发现，ｍＩＬ２＋Ｂ１６ＰＥＬ中的含量明显多于Ｂ１６ＰＥＬ，以上这些说明ｍＩＬ２＋Ｂ１６诱导的ＰＥＬ抗肿瘤能力并非是通过免疫细胞数量上的增加来实现。Peritoneal cavity exudate lymphocytes(PEL)were induced by mIL2 secreting mouse melamoma cell(mIL2+B16)after their inoculation in mouse peritoneal cavity.Have analysed the kinetic change of the mount,components,cytotoxity percent of PEL in different day induced by mIL2+B16 and compared with its parent B16 cells.The results indicate that there are different days.But there are distinct different kinetic change law between two strains PEL.Otherwise in total state,there are no distinct different in the mount level and components of two strains PEL.But the cytotoxity percent of mIL2+B16 PEL is twiceas that of B16PEL.To observe the special granules in lymphocytes cytoplasm,have observed the cytoplasm granules(contains Perforin,Granzymes) in mIL2+B16PEL are more than those in B16 PEL under electron microscope.All above indicates the more antitumor ability of mIL2+ B16 PEL is because their quality have been changed by IL2.
