摘  要:目的:应用飞行时间质谱技术筛选上皮性卵巢癌患者与正常人血清中差异性蛋白,探讨蛋白质组学诊断模型用于上皮性卵巢癌的早期诊断的可行性。方法:应用弱阳离子芯片(WCX2)及表面增强激光解析电离飞行时间质谱仪(SELDI-TOF-MS)检测20例正常人(对照组)和40例上皮性卵巢癌(观察组)患者血清中的差异蛋白质,并检测差异蛋白筛选上皮性卵巢癌的特异性和敏感度。结果:对照组与观察组在1500~20000Da间,质荷比为3865Da、4318Da及5089Da的蛋白峰差异有统计学意义(P<0.05)。3个蛋白区分对照组与观察组的敏感性与特异性为80.00%(32/40)、85.00%(17/20)。结论:SELDI-TOF-MS技术对上皮性卵巢癌早期诊断具有一定价值。Objective To detect the serum markers in epithelial ovarian cancer by SELDI-TOF-MS,and lay a foundation for the early screening. Methods Serum proteins of 20 healthy people and 40 cases with ovarian cancer were detected and compared by WCX2 chip and SELDI-TOF-MS. Results From 1 500 Da to 20 000 Da,the content of three proteins (3 865 Da,4 318 Da) had significant difference in the three groups.The sensitivity and specificity of the three proteins were 80.00% (32/40 and 85.00% (17/20). Conclusion SELDI-TOF-MS technology shows great potentiality for early diagnosis of ovarian cancer and specific tumor marker screening,and deserves generalization.
