摘  要:目的探讨糖基转移酶(GT)催化底物的糖基化反应。方法采用TOPO-TA克隆法建立N-乙酰氨基半乳糖转移酶(ppGalNAcT)、硫酸基(磺基)转移酶(GlcNAc6ST)、半乳糖基转移酶(GalT)和N-乙酰氨基葡萄糖转移酶(GnT)等4类糖基转移酶的cDNA文库,并以此制成表达谱基因点阵。利用此点阵采取化学发光法检测不同肿瘤细胞株中4类糖基转移酶的相应表达谱。结果在不同肿瘤细胞中ppGalNAcT、GlcNAc6ST、GalT和GnT基因的表达水平不一;而在同一肿瘤细胞中,4类糖基转移酶基因的表达也有差异。结论这项研究为以后利用糖基转移酶的表达谱来检测肿瘤的发生、发展提供了依据。Objective To study the reactions of glycosylation by glycosyltransferases. Methods Firstly we established the cDNA libraries of ppGalNAcT,GlcNAc6ST,GalT and GnT by means of TOPO-TA cloning. Using these libraries we created the arrays for gene expression profiling and then they were used to detect different positive signals of ppGalNAcT,GlcNAc6ST,GalT and GnT genes expression in different human tumor cells by chemiluminescent method. Results The results showed that in different human tumor cells,the gene expression levels of the four groups of GT were different from each other. And in each tumor cells,these genes also differed in expression levels. Conclusion This study may provide the basis for detection of tumorigenesis and tumor development in the future.
