联合使用曲格列酮、9-顺维甲酸对胃癌SGC7901细胞细胞周期影响的实验研究

摘  要:目的探讨曲格列酮(TGZ)、9-顺维甲酸(9-cis-RA)联合用药对胃癌SGC7901细胞细胞周期的影响。方法体外培养SGC7901细胞给予TGZ、9-cis-RA干预,采用四甲基偶氮唑蓝(MTT)法、流式细胞术和免疫细胞化学方法检测TGZ、9-GS-RA作用后SGC7901细胞生长情况、细胞周期的改变和p21、p27表达情况。结果MTT结果显示,应用TGZ与9-cis-RA作用于SGC7901细胞72 h,50μmol/L TGZ组、20μmol/L 9-cis-RA组及10μmol/L9-cis-RA+25μmol/L TGZ组细胞的生长受到抑制(P<0.05)。金正均法检测显示两药对SGC7901细胞的抑制有协同作用。免疫细胞化学方法显示50μmol/L TGZ组、20μmol/L 9-cis-RA组及10μmol/L9-cis-RA+25μmol/L TGZ组与阴性对照及低浓度组相比,p21、p27阳性表达率增加(P<0.05)。流式细胞术显示50μmol/L TGZ组、20μmol/L 9-cis-RA组及10μmol/L 9-cis-RA+25μmol/L TGZ组处于G0/G1期细胞增多(P<0.05)。结论TGZ与9-cis-RA可通过诱导肿瘤细胞周期阻滞而发挥抑制胃癌SGC7901细胞增殖的作用,诱导周期阻滞的作用与药物作用后p21、p27表达上调有关。Objective To investigate the effect of troglitazone(TGZ) combined with 9-cis-RA on the cell cycle of gastric carcinoma cell line SGC7901. Methods Human gastric cancer cell line SGC7901 cultured in vitro was treated with TGZ and 9-cis-RA,and then the effect of the growth of SGC7901 cells was detected by MTT assay,the cell cycle of the tumor cells was analyzed by flow cytometry,and the expression of p21 and p27 was detected by immunocytochemistry assays,respectively. Results MTT showed that at the time point of 72 nd hour after the treatment of TGZ combined with 9-cis-RA,the growth of SGC7901 cells was inhibited at the concentration of 50 μmol/L of TGZ,20 μmol/L of 9-cis-RA and 25 μmol/L+10 μmol/L of TGZ combined with 9-cis-RA,respectively(P〈0.05).Determination with Jin Zhengjun method showed that TGZ and 9-cis-RA had synergistic effect on the inhibition of SGC7901 cells.Immunocytochemical method showed that in 50 μmol/L TGZ group,20 μmol/L 9-cis-RA group and 10 μmol/L 9-cis-RA+25 μmol/L TGZ group the positive expression percentage of p21 and p27 increased(P〈0.05),as compared to the negative control and low concentration groups.Flow cytometry showed that in 50 μmol/L TGZ group,20 μmol/L 9-cis-RA group and 10 μmol/L 9-cis-RA + 25 μmol/L TGZ group the cells in G0/G1 phase proliferated(P〈0.05).Conclusions TGZ and 9-cis-RA can inhibit cell proliferation by inducing cell cycle arrest in SGC7901 cells,and the induction of cell cycle arrest is involved in the up-regulation of the expression of p21 and p27.
