摘 要:目的探讨血红素加氧酶-1(HO-1)及其催化产物在远端缺血预适应(RPC)后的心肌梗死大鼠心肌中的作用。方法实验分为心肌梗死组(AMI组)及其假手术组(As组)和下肢缺血预适应组(LIP组)及其假手术组(Ls组)。AMI组和LIP组又分2、4、6、12h组;Ls组实验结束即刻处死;取12h组用伊文思兰和氯化三苯四氮唑(TTC)染色检测心肌梗死面积。用逆转录多聚酶链反应(RT-PCR)检测心肌组织中HO-1 mRNA基因的表达,用全自动生化仪检测血清中的肌酸激酶同工酶(CK-MB)和胆红素。结果LIP组心肌梗死面积比AMI组明显降低(P<0.05);血清中各时间点心肌组织中HO-1的表达、胆红素和铁蛋白均比AMI组明显升高(P<0.05或<0.01)。结论下肢缺血预适应能够减小心肌梗死面积,HO-1对心肌有保护作用。Objective To investigate the role of HO-1 and its catalysis production in distal ischemic preconditioning myocardium of rats with acute myocardial infarction. Methods Healthy male Sprague-Dawley rats were randomly divided into two groups:acute myocardial ischemia group (AMI) and sham operation group;the lower limbs ischemic precondition(LIP) and sham operation group. The AMI and LIP group were divided into 2h,4h,6h,12h group. The rats in sham operation group were killed and whose myocardium tissues were taken after ligating the left anterior descending coronary artery. The myocardial infarction area size was examined with Evans and triphenyl tetrazolium chloride TTC. The expression levels of HO-1mRNA were detected in myocardium by reverse transcription polymerase chain reaction (RT-PCR) method. The levels of TB and CK-Mb in serum were measured with automatic biochemistry analyzer. The quantity of SF in serum was measured by double-antibody 125I radio immunoassay. Results The myocardial infarction area was decreased significantly in LIP group,as compared with that in AMI group;the levels of TB and SF in serum were increased significantly in LIP group ,as compared with that in AMI group atn the same time ,but the activity of CK-Mb in serum was decreased significantly in LIP group,as compared with that in AMI group. The expression of HO-1 mRNA was increased significantly in group LIP,as compared with that in AMI group at the same time. Conclusion The distal ischemic preconditioning of lower limbs can decrease the myocardial infarction area ,and HO-1has protective effect on myocardium.
