磁性纳米簇标记小鼠胚胎干细胞的体外研究

摘  要:通过制备表面带正电荷(Cationic Magnetic Nanoclusters,CMNCs)和负电荷(Anionic Magnetic Nanoclusters,AMNCs)的磁性纳米簇,分别在体外对小鼠胚胎干细胞(mouse Embryonic Stem Cells,mESC)进行标记.采用Prussianblue染色、电镜观察鉴定2种磁性纳米簇对mESC标记率的结果表明:CMNCs细胞标记组铁浓度在0.03~0.09mmol/L和AMNCs细胞标记组铁浓度在0.3~0.9mmol/L时,90%以上细胞被标记,透射电镜可见磁性纳米簇颗粒位于胞浆中.用7-AAD和CCK-8试剂盒分别检测标记后mESC的活力和增殖能力,结果显示:CMNCs细胞标记组在低于0.3mmol/L浓度标记后,对mESC的增殖能力与正常未标记组比较,差异无统计学意义(P〉0.01);AMNCs细胞标记组在高于0.06mmol/L浓度标记时与对照组比较,差异有统计学意义(P〈0.01).与AMNCs相比,CMNCs在mESC标记方面表现出了优良的特性.Cationic magnetic nanoclusters (CMNCs) and anionic magnetic nanoclusters (AMNCs) were prepared, and then were used to label mouse embryonic stem cells (mESC) in vitro. Prussian blue staining and electron microscopy were used to evaluate the labeling efficiency of these two kinds of nanoclusters to mESC: when the concentration of CMNCs is 0. 03--0. 09 mmol/L and the concentration of AMNCs is 0.3-0.9 retool/L, it shows that more than 90% of mESC are positively labeled and magnetic nanoclusters are found in their cytoplasm by transmission electron microscopy. 7-AAD and CCK-8 kits were used to assess the ability of labeled mESC to survive and proliferate, respectively. The results show: when the concentration of CMNCs is less than 0.3 mmol/L, there is no significant difference between labeled and unlabeled mESC in their abilities to proliferate (P〉0.01). However, when the concentration of AMNCs is higher than 0.06 mmol/L, there is significant difference between labeled and unlabeled mESC in their abilities to proliferate (P〈0.01). So CMNCs is an efficient agent for labeling mESC and is superior to AMNCs.
