摘 要:目的观察牛磺酸对甲基汞致大鼠脑氧化损伤的影响。方法24只大鼠,分成3组,每组8只。第1组为对照组,第2组单纯染甲基汞组,第3组为牛磺酸预处理组。第1、2组皮下注射0.9%氯化钠溶液,第3组皮下注射2mmol/kg的牛磺酸。2 h后,第1组腹腔注射0.9%氯化钠溶液,第2、3组腹腔注射25μmol/kg的氯化甲基汞溶液。染毒后24 h处死大鼠,切取大脑皮质,测定大鼠大脑皮质汞含量,谷胱甘肽(GSH)和丙二醛(MDA)含量,谷胱甘肽过氧化物酶(GSH-Px)和超氧化物歧化酶(SOD)活力。结果染甲基汞24 h后,与对照组比较,单纯染甲基汞组大鼠大脑皮质汞含量显著升高(P<0.01),GSH含量显著降低(P<0.01),MDA含量明显升高(P<0.05),而GSH-Px和SOD活力均明显下降(P<0.05);与单纯染甲基汞组比较,牛磺酸干预组大鼠大脑皮质汞含量差异无统计学意义(P>0.05),GSH含量明显升高(P<0.05)和MDA含量明显下降(P<0.05),而GSH-Px活力显著回升(P<0.01)和SOD活力明显回升(P<0.05)。结论牛磺酸对急性甲基汞致大鼠脑氧化损伤有一定拮抗作用。Objective To determine the effect of taurine on oxidative injure induced by methylmeroury on cerebrum in rats. Methods Twenty- four Wistar rats were randomly divided into three groups by weight. The first group was the control group, the second was MeHg- exposed group, and both were given 0.9% NaCl. The third group was subcutaneously injected with 2 mmol/kg of taurine. Two hours later, the control group was intraperitonally given 0.9% NaCl, the second and the third groups were intraperitonally given 25 μmol/kg of methylmercury chloride. The volume of injection was all 5 ml/kg. 24 hours later, the rats were killed and the cerebral cortices were taken for detecting the contents of Hg, GSH and MDA and the activities of GSH- Px and SOD in pallium. Results As compared with the control group, the content of Hg and MDA of the MeHg - exposed group were increased significantly, the activity of GSH - Px, SOD and the content of GSH of the MeHg - exposed group were decreased significantly. As compared with the MeHg - exposed group, the content of MDA of the taurine treated group was decreased significantly, the activity of GSH- Px, SOD and the content of GSH of the taurine treated group were increased significantly. No statistically significant difference was found in the content of Hg between the MeHg- exposed group end the taurine treated group (P〉0.05). Conclusions Methylmercury can cause oxidative injure in rats, and taurine can effectively counteract the effect of methylmercury.
