摘 要:目的观察罗格列酮对肝星状细胞增殖与Ⅰ型胶原合成的影响。方法采用HSC-T6肝星状细胞系,将培养的肝星状细胞(HSC)分为对照组、不同浓度的罗格列酮组(5、10、20μmol/L)。采用MTT法观察罗格列酮对HSC增殖的影响,采用ELISA法检测细胞培养液上清中Ⅰ型胶原的含量。结果与对照组相比,在10、20μmol/L罗格列酮组,罗格列酮可以抑制HSC的增殖(P<0.05),同时细胞培养液上清中I型胶原含量分别为135.72±6.00ng/ml、128.65±5.34ng/ml,均低于对照组(149.35±6.65ng/ml),差异有统计学意义(P<0.05)。结论罗格列酮可以抑制HSC的增殖,抑制肝星状细胞I型胶原的合成。Objective To investigate the effects of rosiglitazone on the proliferation and collagen Ⅰ synthesis of cultured hepatic stellate cells(HSC) in vitro. Methods HSC line, HSC-T6 was incubated with different concentraion of rosiglitazone(5, 10,20 μmol/L). Cell proliferaeion was examined by MTT colormetric assay. Cell culture medium was used to detect collagen Ⅰ level with ELISA method. Results Compared to control group, 10,20 μmol/L rosiglitazone groups can inhibit HSC proliferation ( P 〈0. 05). The collagen Ⅰ level of 10,20 μmol/L rosiglitazone groups were 135.72 ±6. 00 ng/ml,128.65 ±5.34 ng/ml respectively, which were significantly lower than control group( 149. 35 ±6. 65 ng/ml, P 〈 0. 05 ). Conclusion Rosiglitazone can inhibit HSC proliferation, decrease the collagen Ⅰ synthesis.
