摘  要:目的:本研究对8种人结肠癌细胞系的干细胞特性进行初步检测,以寻找适合分选结肠癌干细胞的细胞系。方法:利用流式细胞术,RT-PCR检测8种结肠癌细胞中CD133的表达情况;利用条件培养观察8种细胞系的肿瘤细胞在无血清培养基中的生物学特性;利用裸鼠成瘤模型检测成瘤能力和转移能力。结果:CD133在8种细胞系中的表达有显著差异,最高的HCT116细胞系中CD133+细胞含量为(91.4±5.0)%;各种细胞系细胞均能在无血清培养基中增殖,但只有HCT116细胞能形成典型的"神经球样结构"且具有较高的成瘤能力和转移能力。同等细胞浓度下移植瘤体积HCT116(1.06±0.13)cm3,LOVO(0.22±0.09)cm3,P=0.01,差异具有统计学意义。结论:相对于其他7种细胞系,HCT116细胞具有多种肿瘤干细胞的生物学特性,更适合于结肠癌干细胞的研究。OBJECTIVE:To investigate the characteristics of colon cancer stem cells in colon cancer cell lines and look for suitable cell lines applying to isolate colon cancer stem cells.METHODS:The expressions of CD133 in eight cell lines were investigated by flow cytometry and RT-PCR.The different cancer cells were cultured in serum-free medium for the study of their Biological characteristics.The tumor model in nude mice was used to evaluate the tumorigenicity and metastasis of different cancer cells.RESULTS:The expressions of CD133 in eight cell lines were significantly different and the highest HCT116 cell line contained(91.4±5.0)% CD133+ cells.Eight types of cancer cells could proliferate in serum-free medium,but only HCT116 cells could form the typical "neurosphere" structure and had high tumorigenicity.At the same cell density,the xenografts formed by HCT116 cells(1.06±0.13)cm3 were statistically bigger than that of LOVO cells(0.22±0.09)cm3,P=0.01.CONCLUSION:Compared with other seven cell lines,HCT116 possesses the main defining characteristics of colon cancer stem cells,so it could be a suitable cell line for researches of colon cancer stem cells.
