摘  要:目的研究人脑出血后神经生长因子前体(proNGF)、sortilin在血肿周围组织中的表达及与细胞凋亡的关系。方法本实验采集临床手术患者的脑组织标本,利用末端脱氧核苷酸转移酶介导的脱氧尿苷三磷酸生物素原位缺口末端标记法(TUNEL)检测血肿周围组织中的凋亡细胞,运用免疫组化技术检测血肿周围组织及远隔部位组织中proNGF、sortilin的表达。结果脑出血后血肿周围组织中细胞凋亡率明显增高(P<0.01),proNGF的表达水平并没有显著变化(P>0.05),而sortilin的表达水平则明显增加(P<0.01),并且sortilin的表达水平与细胞凋亡率正相关(R=0.648,P=0.00)。结论sortilin在人脑出血后表达增加,与细胞凋亡正相关,proNGF在脑出血后的表达量并未变化。Objective To investigate the expression of pro-form of nerve growth factor (proNGF) and sortilin in brain tissue surrounding hematoma after intracerebral hemorrhage (ICH) and its relation to apoptosis. Methods In this study, the brain tissue samples were collected during operation and investigated with terminal deoxynucleotidyl transferase mediated deoxyuridine triphosphate nick end labelling (TUNEL) assay to detect apoptotic cells. Meanwhile, the protein expression of proNGF and sortilin was detected with immunohistochemistry in brain tissue surrounding and distant to the hematoma respectively. Results After ICH apoptosis increased significantly in the surrounding tissues of hematoma ( P 〈 0. 01 ). The expression level of sortilin increased significantly (P 〈0. 01 ), but the expression level of proNGF did not change greatly (P 〉0.05). And there was a statistical significant correlation between sortilin expression and TUNEL positive cells ( R = 0. 648, P = 0.00). Conclusion Expression of sortilin increases after ICH, which is positively correlated with apoptosis, while expression of proNGF does not change after ICH.
