摘 要:目的:观察不同浓度五倍子水提取物对内毒素抑制人牙髓细胞碱性磷酸酶活性的影响。方法:采用组织块法体外培养人牙髓细胞,以含不同浓度五倍子水提取物2.5、5、10、20、40、100μg/mL内毒素和20 mL/L胎牛血清的DMEM共同培养第5代人牙髓细胞,对照组用只含100μg/mL LPS、200 mL/L胎牛血清的DMEM培养。分别于培养后1、2、3、4、5 d,用酶联免疫检测仪检测细胞碱性磷酸酶活性,采用单因素方差分析(完全随机设计)、t检验对实验数据进行统计学分析。结果:2.5、5、10μg/mL五倍子水提取物均能明显拮抗100μg/mL内毒素对人牙髓细胞碱性磷酸酶活性的抑制作用,其中以5μg/mL五倍子水提取物拮抗作用最好,且随时间延长,拮抗作用越明显。20μg/mL和40μg/mL五倍子水提取物拮抗LPS的作用不明显。结论:低浓度的五倍子水提取物具有拮抗内毒素对人牙髓细胞碱性磷酸酶活性的抑制作用,并呈时间依赖性。AIM: To observe the influence of Chinese nutgall extracts on the inhibitory effect of lipopolysaccharide to ALP activity of human dental pulp cells in different doses and action time. METHODS: Human dental pulp ceils were cultivated by tissue explant. Human dental pulp cells of the fifth passage were cultured in DMEM with 100 p^g/mL lipopolysaecharide, Chinese nutgall extracts(2.5 to 40p.g/mL) and 20mL/L fetal bovine serium. ALP activity of cells was analyzed. One-way ANOVA( completely random design)and t -test were used in statistical analysis. RESULTS: Low -dose of Chinese nutgall extracts( ≤ 10 μg/mL) can resist the inhibitory effect of lipopolysac- eharide to ALP activity of human dental pulp cells significantly. The 5 μg/mL is the best. From the second day , the ALP activity of human plup cells incubated with 5μg/mL Chinese nutgall extracts and 100p, g/mL lipopolysaccharide increased signicificantly in a time - dependent manner. The action of 20 p^g/mL and 40 p^g/mL is not obvious. CONCLUSION : Low - dose Chinese nutgall extracts can resist the inhibitory effect of lipopolysaccharide to ALP activity of human dental pulp cells.
