三羟异黄酮对孕期乳腺癌(MMTV)-ErbB-2转基因子代小鼠乳腺肿瘤的干预作用

摘　　要：目的观察三羟异黄酮干预孕期乳腺癌(MMTV)-ErbB-2转基因小鼠对其子代小鼠乳腺肿瘤发生、发展的影响。方法按三羟异黄酮20μg、100μg溶于2%二甲基亚砜(DMSO)和0.05mL植物油中,对照组给予等量植物油处理,将孕鼠随机分为低、高剂量三羟异黄酮给药组和正常对照组,每组30只,自孕期第13～19天采用母鼠后腿外侧皮下注射,观察三羟异黄酮给药组子代雌性小鼠和对照组子代雌性小鼠乳腺肿瘤的潜伏期和乳腺成瘤率是否存在差异,并用免疫组织化学染色SP方法检测子代小鼠乳腺肿瘤组织p53的表达情况。结果高剂量组乳腺肿瘤潜伏期、乳腺成瘤率与其他各组相比具有统计学差异(P<0.05);高剂量组p53的阳性表达水平明显低于其他各组(P<0.05)。结论孕期使用高剂量异黄酮可以延长子代雌性小鼠肿瘤发生的潜伏期,降低乳腺成瘤率,并且可以降低子代小鼠乳腺肿瘤中p53的阳性表达水平,提示三羟异黄酮对乳腺癌(MMTV)-ErbB-2转基因小鼠乳腺肿瘤的发生、发展具有一定的预防作用。Objective Observation of genistein interferes with breast cancer during pregnancy （MMTV）-ErbB-2 transgenic mice to their offspring mouse mammary tumor occurrence and development impact.Methods According to genistein 20μg,100μg dissolved in 2% DMSO （dimethyl sulfoxide）,and 0.05ml vegetable oil in the control group was given equal amount of vegetable oil processing.The pregnant rats were randomly divided into low and high dose of genistein treatment group and normal control group,n=30,since the pregnancy the first 13 to 19 days using female mouse hind leg lateral subcutaneous.Observation of genistein treatment group were offspring of female offspring of female mice and the control group mouse mammary tumor latency and the existence of mammary tumor formation rate differences,and SP immunohistochemical staining to detect breast tumors in mice offspring p53 expression.Results The incubation period of high-dose group of breast cancer,breast tumor formation rate compared to other groups with significant difference （P〈0.05）;high-dose group p53 positive expression was significantly lower than other groups （P〈0.05）.Conclusion The use of high doses of isoflavones during pregnancy can be extended offspring of female mice with tumor latency,reducing the rate of breast tumor formation,and may reduce the offspring of mice with breast tumor levels of positive expression of p53,suggesting that genistein on breast cancer （MMTV）-ErbB-2 transgenic mouse mammary tumor occurrence and development of a certain preventive effect.
