摘 要:目的:为了检测细胞间隙连接蛋白基因家族中哪些成员在鼻咽癌细胞中表达,研究分化诱导剂对这些基因的表达作用和机制。方法:细胞生长曲线,Northern杂交。结果:细胞间隙连接蛋白基因(connexin,Cx)家族成员的两个基因:Cx37、Cx46在鼻咽癌细胞株HNE1表达。维甲酸能抑制HNE1的生长和Cx37和Cx46在HNE1中的表达,并能诱导RARa的表达。去除维甲酸处理后细胞生长抑制被解除,Cx37、Cx46、RARa的表达恢复正常。结论:Cx37和Cx46是鼻咽癌细胞中特异表达的Cx家族基因,维甲酸能抑制Cx46和Cx37的表达,可能的机制是通过诱导RARa的表达而起作用,维甲酸对Cx基因表达调节是可逆的。Purpose:To detect which members in connexin gene family are expressed in nasopharyngeal carcinoma cell line HNE1,and the mechanism by which those genes are specifically switched on and off during retinoic acid (RA)induction. Methods:Cell growth curves,Northern hybridization were used. Result:Two genes:Cx46 and Cx37 which belong to connexin gene family,were expressed in HNE1 The down regulation of Cx46 and Cx37,up regulation of RARa and growth inhibition were observed in HNE1 after exposure to RA.Furthermore,the gene expression and cell growth in HNE1 cell were restored after removal of RA.Conclusion:2 members of connexin gene family:Cx37 and Cx46 were expressed in HNE1 cell,RA can inhibit the expression of these two genes mediated by RARa,the effects of RA on HNE1 are reversible.
