摘 要:目的:建立以反相高效液相色谱法测定大鼠血浆及胃肠道内容物、黏膜中酮洛芬浓度的方法。方法:色谱柱为Hypersil C18,流动相为甲醇-50mmol·L-1磷酸二氢钾缓冲液(pH3.5,52∶48,V/V),检测波长为258nm,柱温为30℃,流速为1mL·min-1。结果:酮洛芬在各生物样本中检测浓度范围内线性关系良好(r为0.9926~0.9999);平均绝对回收率为83.76%~99.43%,平均方法回收率为86.41%~115.68%;日内RSD小于9.85%,日间RSD小于14.84%。结论:本方法准确、快速、灵敏,可用于生物样品中酮洛芬的浓度测定。OBJECTIVE: To establish an RP-HPLC method for the determination of plasma, gastrointestinal content and mucosa concentration of ketoprofen in rats. METHODS: The separation was performed on Hypersil C18 chromatographic column. The mo bile phase consisted of methanol-50 mmol.L^-1 potassium dihydrogen phosphate buffer (pH 3.5,52:48, V/V) with a detection wavelength of 258 nm. The column temperature was 30℃ and the flow rate was 1 mL.min^-1. RESULTS: Good linear relationships were achieved when the detection concentrations of ketoprofen were within the range (r=0.992 6-0.999 9). The average absolute recovery rate was 83.76%-99.43%, the average methodological recovery rate was 86.41%-115.68%, the intra-day RSD was less than 9.85% and the inter-day RSD was less than 14.84%. CONCLUSION: This method is accurate, rapid, sensitive and satisfying enough to be applied to measure the concentration of ketoprofen in biological samples.
