央芪汤对胃癌细胞凋亡影响的实验研究

摘　　要：目的观察央芪汤对胃癌细胞凋亡的影响。方法培养人胃癌细胞SGC-7901细胞株,用MTT法评价央芪汤对胃癌细胞增殖的抑制作用,确定央芪汤对胃癌细胞的IC50值,采用流式细胞术检测央芪汤对胃癌细胞凋亡的诱导作用,免疫细胞化学法检测胃癌细胞中bcl-2及caspase-3蛋白的表达情况。结果央芪汤可以显著促进胃癌细胞SGC-7901的凋亡(P<0.01),明显增加胃癌细胞SGC-7901凋亡蛋白caspase-3的表达(P<0.01),并可明显抑制抗凋亡蛋白bcl-2的表达(P<0.01)。结论央芪汤可以通过促进胃癌细胞的凋亡,达到抗肿瘤的目的。Objective It is to observe the influence of gastric cancer cell apoptosis treated with Yangqi decoction.Methods The type SGC-7901 of gastric cancer cell was bred,the inhibitory effect of Yangqi decoction on gastric cancer cell increment by MTT,and the IC50 of Yangqi decoction was determined;the induction of Yangqi decoction on gastric cancer cell were detected by flow cytometry;the expressions of bcl-2 and caspase-3 protein in gastric cancer cell were detected by immunocytochemistry.Results The effect of gastric cancer cell apoptosis with Yangqi decoction were improved obviously,the expression of SGC-7901-apoptotic protein caspase-3 in gastric cancer cell was significantly increased and the expression of anti-apoptotic protein bcl-2 was significantly inhibited.Conclusion Yangqi decoction can obviously improve gastric cancer cell apoptosis to achieve the purpose of anti-tumor.
