摘 要:目的:探讨活性氧(reactive oxygen species,ROS)改变在PIG11蛋白高表达诱导人肝癌HepG2细胞凋亡中的作用。方法:本研究采用2’,7’-二氯氢化荧光素二脂(2’,7’-dichlorofluorescin diacetate,DCFH-DA)荧光探针标记PIG11蛋白高表达肝癌细胞株pLXSN-PIG11-HepG2细胞内ROS,用N-乙酰半胱氨酸(N-acetylcystine,NAC)清除细胞内ROS后,FCM检测细胞内ROS水平的变化以及细胞凋亡率。结果:FCM测定结果显示,pLXSN-PIG11-HepG2细胞内ROS水平(15.60±0.92)明显高于pLXSN-HepG2细胞(4.90±0.70)和HepG2细胞(5.37±1.17),差异有统计学意义(P<0.01)。NAC(10 mmol/L)清除细胞内ROS后,FCM检测显示pLXSN-PIG11-HepG2细胞凋亡率较未清除前明显降低(P<0.01)。结论:PIG11高表达诱导细胞凋亡的机制可能与细胞内ROS水平升高有关。Objective:To investigate the role of reactive oxygen species(ROS) in the apoptosis of human hepatocellular carcinoma HepG2 cells induced by PIG11 protein overexpression.Methods:The fluorescent probe 2',7'-dichlorofluorescin diacetate(DCFH-DA) was used to label the intracellular ROS in pLXSN-PIG11-HepG2 cells,pLXSN-HepG2 cells and HepG2 cells.The intracellular fluorescence intensity was detected by using the flow cytometry(FCM).The apoptotic ratio of HepG2was determined by FCM after elimination of intracellular ROS with 10 mmol/L N-acetylcystine(NAC).Results:The intracellular content of ROS in pLXSN-PIG11-HepG2 cells was significantly higher than that in pLXSN-HepG2 cells[(15.60±0.92) vs(4.90±0.70),P0.01)].The apoptotic ratio of pLXSN-PIG11-HepG2 cells was significantly decreased by pretreatment with 10 mmol/L NAC(P0.01).Conclusion:The apoptosis-inducing effect of PIG11 over-expression is related with elevation of intracellular ROS levels.
