依达拉奉对大鼠局灶性脑梗死Caspase-3表达的影响

摘　　要：目的探讨依达拉奉对大鼠局灶性脑梗死Caspase-3表达的影响，明确依达拉奉的抗调亡机制。方法将Wistar大鼠分为依达拉奉治疗组、脑梗死对照组和假手术组，于14d采用RT-PCR进行Caspase-3 mRNA检测，采用Western blot方法进行蛋白检洲，并进行组间比较。结果脑梗死对照组Caspase-3 mRNA及其蛋白的表达量较假手术组增加；经过依达拉奉治疗后，两指标的表达最有所降低（P〈0．01）。结论依达拉奉可以减少Caspase-3 mRNA及其蛋白的表达，发挥其神经细胞保护作用。Objective To investigate effect of Edaravone on Caspase-3 expression in the rats with Local cerebral infarction and to determine anti-apoptosis mechanism ofEdaravone.Methods Wistar rats were divided into Edaravone treatment group, cerebral infarction group and sham-operated group.Caspase-3 mRNA expression was detected by RT-PCR, Caspase-3 protein expression was assessed by Western blot.Results Expression of Caspase-3 mRNA and protein in cerebral infarction group were more than those in sham-operated group, but above index in Edaravone treatment group＇ were less than those in cerebral infarction group （P〈0.01）.Conclusion Edaravone can decrease expression of Caspase-3 mRNA＇and protein to play an important role in neuroprotection.
