E.tenella端粒酶逆转录酶基因3′端序列的克隆及分析

摘　　要：为了获得E.tenellaTERT全长cDNA序列,本试验根据已报道的其他,原虫TERT氨基酸序列,利用CODEHOP方法设计简并引物,同时利用Trizol法提取总RNA并反转录为cDNA,然后采用3-′RACE方法克隆获得E.tenellaTERT 3′cDNA序列,并采用生物信息学技术进行序列分析。结果表明,该序列具有TERT蛋白家族特征性基序,证明成功克隆了E.tenellaTERT 3′cDNA序列。In order to get the full length cDNA sequence of E.tenella TERT,degenerate primer was designed by CODEHOP according to the amino acid sequence of other TERTs reported.Total RNA was extracted by Trizol method and reverse transcripted.Then obtained 3′end cDNA of E.tenella TERT gene by 3′-RACE and analyzed the characteristic of sequence by bioinformatics software also.The results of sequence analysis indicated that the sequence had the distinctive motif of TERT protein family.It identified that 3′ end cDNA of E. tenella sequence was cloned successfully in this study.
