光损伤实验性大鼠视网膜组织中survivin表达的变化

摘　　要：目的研究光损伤视网膜组织中survivin表达的变化及其意义。方法将30只Sprague-Dawley大鼠随机分为对照组5只和光损伤组25只。光损伤组分别按照光照后1h、6h、12h、24h、48h分成5个亚组,每组5只大鼠,并于上述5个时间点处死。利用链霉素-生物素-过氧化物酶免疫组织化学法和逆转录聚合酶链反应检测不同时段survivin蛋白和survivin mRNA在视网膜组织表达的变化。结果免疫组织化学染色:对照组视网膜节细胞层、内丛状层、内核层都有survivin蛋白的阳性表达,而光损伤组随着光损伤时间的延长,survivin蛋白阳性表达量逐渐减少,染色由棕黄色变为黄色。对照组灰度值为166.520±4.342,光损伤1h、6h、12h、24h、48h亚组灰度值分别为165.520±1.274、171.330±1.432、176.210±2.464、182.310±2.310、182.920±3.976;survivin mRNA的表达对照组为0.575±0.047,光损伤1h、6h、12h、24h、48h分别为0.563±0.055、0.505±0.024、0.430±0.015、0.368±0.014、0.365±0.027;除光损伤1h亚组与对照组、光损伤24h与48h亚组间survivin mRNA表达和survivin蛋白表达差异无统计学意义(均为P>0.05)外,其余各时间点组间比较及与对照组比较差异均有统计学意义(均为P<0.05)。结论光损伤视网膜组织中survivin蛋白表达降低,并在一定时间具有时间依赖性。Objective To investigate the changes of survivin expression in retina of experimental rats with light injury.Methods Thirty Sprague-Dawley rats were randomly divided into control group （5 rats） and light injury group （25 rats）.The light injury group was divided into 5 subgroups according to light exposure for 1 hour,6 hours,12 hours,24 hours and 48 hours,5 rats in each subgroup,and sacrificed at 5 time points.The expression changes of survivin protein were measured by immunohistochemistry technique.The survivin mRNA was studied by RT-PCR.Results Immunohistochemical staining showed that positive expression of survivin protein was seen at layer of retinal ganglion cells,inner plexiform layer and inner nuclear layer in control group;While the positive expression of survivin protein was gradually decreased and the staining color changed from brown to yellow with prolonging of light injury in light injury groups.Grey values were 166.520±4.342,165.520±1.274,171.330±1.432,176.210±2.464,182.310±2.310 and 182.920±3.976 in control group,light injury for 1 hour,6 hours,12 hours,24 hours and 48 hours groups,respectively.Survivin mRNA expressions were 0.575±0.047.0.563±0.055,0.505±0.024,0.430±0.015,0.368±0.014 and 0.365±0.027 in control group,light injury for 1 hour,6 hours,12 hours,24 hours and 48 hours groups,respectively.No significant difference was found in expression of survivin protein between light injury for 1 hour group and control group or between light injury for 24 hours group and light injury for 48 hours group（both P〈0.05）,while there was statistical difference among different light injury groups or each light injury group compared with control group（all P〉0.05）.Conclusion The expression of survivin protein decreases in retinal tissues of light injury and be in time-dependent manner.
