摘　　要：目的分析结膜松弛症泪液蛋白表达谱的变化。方法收集22例结膜松弛症患者和13例正常对照组的泪液样本,毛细管从每眼收集15μL泪液,采用ZipTip C18柱浓缩和纯化泪液样本,利用基质辅助的激光解吸质谱进行线性模式和肽质量指纹谱(PMF)检测,并对PMF图谱数据进行多变量数据统计分析,比较结膜松弛症组和正常对照组的泪液PMF的差异。结果2组泪液的质谱检测峰在数量上差异无统计学意义(P>0.05),主成分分析结合偏最小二乘法分析显示结膜松弛症组泪液蛋白质表达与正常对照组比较有差异,结膜松弛症组泪液蛋白质表现为"离群"样本,且离群分散度较大的样本为结膜松弛症Ⅳ级的患者。结论应用ZipTipC18柱分离联合基质辅助的激光解吸质谱技术可有效筛查泪液蛋白质。对PMF图谱进行多变量数据统计分析,可以观察到结膜松弛症组与正常对照组泪液蛋白质存在差异,并可为结膜松弛症临床诊断、疾病发生发展的机理及治疗提供参考。Objective To analyze the changes of tear proteins expression in conjunctivochalasis.Methods Tear samples were obtained from 22 conjunctivochalasis cases and 13 normal controls.Capillary tube was used to collect 15 μL tear from each eye.Tear samples were concentrated and purified with ZipTip C18 column.Matrix assisted laser desorption/ionization time-of-flight mass spectrometry（MALDI-TOF-MS） was used for linear model and peptide mass fingerprinting（PMF） analysis.PMF data were analyzed by multivariate statistical techniques to compare the differences of tear PMF between conjunctivochalasis and normal controls.Results The difference between conjunctivochalasis and normal controls according to the peak number of mass spectra was not significant（P〉0.05）.Principal component analysis with partial least squares regression analysis showed there were differences of tear proteins expression between conjunctivochalasis and normal controls.The tear proteins from conjunctivochalasis were strayed from normal controls.The samples who strayed more were from conjunctivochalasis patients at Ⅳ level.Conclusion ZipTip C18 column combined with MALDI-TOF-MS can effectively screen tear proteins.The PMF data can be analyzed by multivariate statistical techniques,which shows differences of tear proteins between conjunctivochalasis and normal controls,and provides references for clinical diagnosis,pathogenesis and treatment of conjunctivochalasis.
