摘 要:多源耐药基因的过度表达是导致白血病化学药物治疗失败的主要原因之一。在正常细胞组织中多源耐药基因的功能是使编码细胞膜蛋白,防止脂类物质在细胞中的堆积及甾体激素的转运。由于细胞膜蛋白及其mRNA在细胞中含量极低,应用传统的蛋白质检测手段及Northern blot检测其水平不够敏感。本研究建立了应用逆转录PCR技术对多源耐药基因的定量检测,其方法灵敏、快速,具有极大的临床应用价值。An excessive expression of multidrug resistance gene is recently considered to be one of the reasons that lead to the failure of chemotherapy probably in childhood leukemias. The major functions of this gene in normal cells is to code protein of cell membrane, and prevent from the accumulation of lipids in cells and the transportation of steroid hormones. It would be less sensitivity using triditional assays of both membrane protein and Northern blot owing to very low quantity of mambrane protein and its mRNA in cells. The present study set up a sensitive and rapid RT-PCR technique for determining multidrug resistance gene quantitatively, it is siginificant to have great valuable in clinical practice.
