细胞甩片、点片和涂片法原位杂交检测急性白血病多药耐药基因表达

摘  要:目的:研究不同取材方法对检测白血病细胞多药耐药(MDR1)基因表达的意义。方法:用细胞甩片、点片和涂片三种取材方法做原位杂交,检测了10例急性白血病MDR1基因表达水平。结果:发现10例中7例表达MDR1基因,MDR1基因阳性率甩片法和点片法较高,涂片法较低;细胞形态学以甩片和涂片法较好,而点片法较差。结论:三种方法均可用于MDR1基因检测,其中甩片法较可靠。Objective:To compare the results of different sampling methods in detecting multidrug resistance (MDR1) gene expression in acute leukemia cells. Methods:The MDR1 gene expression in acute leukemia cells from 10 patients were detected by in situ hybridization after the cells were spun, dripped and smeared on the sheet gasses. Results:The MDR1 mRNA was revealed in the cells from 7 of 10 leukemia patients. It was found that the positive rates of MDR1 gene expression in the spun cells were similar as dripped cells, but higher than smeared cells. The morphologic features of spun and dripped cells were much better than smeared cells.Conclusion:All three sampling methods(spin, drip and smear) may be used to detect MDR1 gene expression in leukemia cells. And the method of cytospins seems to be better than the methods of cell drip and cells smear.
