经鼻给小剂量重组人促红细胞生成素对大鼠癫痫的影响

摘　　要：目的:研究经鼻给小剂量重组人促红细胞生成素(r-HuEPO)对大鼠癫痫发作的影响。方法:大鼠经鼻或腹腔给r-HuEPO后,观察对戊四唑(PTZ)诱导癫痫发作潜伏时间、级别、大发作持续时间、次数的影响。另外,大鼠经鼻给r-HuEPO后,观察对最大电休克(MES)诱导的惊厥发作级别,前肢或后肢强直性伸展时间的影响;对杏仁核电点燃后阈刺激诱导的行为等级,大发作持续时间及后放电的影响。结果:经鼻给予小剂量r-HuEPO,可明显延长PTZ诱导的癫痫发作潜伏时间,降低癫痫发作级别、大发作持续时间和发作次数,其中2.4 IU剂量组作用最为明显。而腹腔给r-HuEPO(3000、4000 IU/kg),仅可减少大发作次数和缩短大发作持续时间。经鼻给2.4 IU的r-HuEPO,可显著降低MES诱导的大鼠惊厥发作级别,缩短前肢和后肢强直性伸展时间。另外,经鼻给2.4 IU r-HuEPO,明显降低杏仁核电点燃后阈刺激诱导的癫痫发作级别,缩短大发作时间和后放电时间。结论:经鼻给小剂量的r-HuEPO有明显的抗癫痫作用。Objective： To investigate the effect of intranasal administration of low dosage recombinant human erythropoietin （r-HuEPO） on seizure intraperitoneal administration of r HuEPO, the behaviora were observed in pentylenetetrazol （PTZ） and maximal electrical amygdaloid-kindled seizure of rats. Results： r-HuEPO increased the seizure latency, and decreased in rats. Methods： After intranasal or and electroencephalographic changes electroshock （MES） induced seizure or Intranasal administration of low dosage the seizure grade and duration, and the number of convulsive episodes in PTZ induced seizure,with the most potential dosage of 2.4 IU. Intraperitoneal administration of r-HuEPO （3 000, 4 000 IU/kg） only decreased the seizure duration and number of convulsive episodes. The seizure grade, forelimb or hindlimb extension duration were decreased in MES-induced seizure by intranasal administration of 2.4 IU r -HuEPO. In addition,intranasal administration of 2.4 IU r-HuEPO decreased the seizure grade, generalized seizure duration and afterdischarge in electrical amygdaloid-kindled rats stimulated with generalized seizure threshold. Conclusion： Intranasal administration of low dosage r-HuEPO can inhibit the seizure in rats.
