摘  要:目的研究洛泊对体外培养的浆液性卵巢癌细胞SKOV3凋亡作用及探讨其对凋亡基因bcl-2和bax表达的影响。方法体外培养卵巢癌细胞SKOV3,用MTT比色法检测洛铂对SKOV3细胞株的增殖抑制作用以及流式细胞仪检测洛铂对SKOV3细胞株的凋亡率,流式细胞仪检测凋亡相关蛋白bax、bcl-2的表达。结果体外培养卵巢癌SKOV3细胞经不同浓度洛泊处理后细胞生长明显受到抑制,洛泊作用于卵巢癌SKOV3细胞可诱导细胞的凋亡,同时,随着洛铂浓度的增加,bax基因的表达增加,而bcl-2表达减少。结论卵巢癌SKOV3细胞对洛泊具有药物敏感性,洛泊可诱导卵巢癌细胞的凋亡,bax蛋白的表达增加和bcl-2蛋白表达的减少可能是洛泊诱导卵巢癌细胞的凋亡机制之一。Objective To investigate the effect of lobaplatin on apoptosis of ovarian cell and its relationship with the expression of bax and bcl-2 gene. Methods MTT assay was used to detect the effect of lobaplatin on proliferation,and apoptosis of SKOV3 ceils were detectrd by flow cytometry.The expression of bax and bcl-2 protein were detected by flow cytometry and Results I_obaplatin could inhibit the proliferation of SKOV3 cells significantly. After treatment with lobaplatin, SKOV3 cells shown some typical morphologic features of apoptosis, the expression of bcl-2 decreased but the bax increased along with the increase of lobaplatin concentration. Conclursion Lobaplatin can inhibit the growth of ovarian cells SKOV3 and induce apoptosis and the down-regulation of bcl-2 and up-regulation of bax many be one of the mechanisms of the apoptosis.
